چکیده

Microtubules are structural and motile elements. They are essential in numerous cell processes. Their function will be greatly improved by our understanding of their molecular structure, which requires extracting microtubules from the Cellular Electron Tomography Images. Manual segmentation and measurement of the microtubules from 3D dataset may require several man-hours of work, and the manual approach suffers from inter-observer and intraobserver variabilities. Localizing microtubules axes is an important step for the successful segmentation. In this paper, a graph searching based technique for extracting central axes of microtubules from three dimensional cellular electron tomography images is presented. A tube enhancement filter is introduced in order to define a distance metric for graph searching. The results are promising, indicating that accurate measurements can be obtained in a semi-automatic fashion.

3D structure of eukaryotic flagella in a quiescent state revealed by cryo-electron tomography.

We have used cryo-electron tomography to investigate the 3D structure and macromolecular organization of intact, frozen-hydrated sea urchin sperm flagella in a quiescent state. The tomographic reconstructions provide information at a resolution better than 6 nm about the in situ arrangements of macromolecules that are key for flagellar motility. We have visualized the heptameric rings of the mo...

Feature Extraction for Analysis of Electron Microscopy Images

Transmission electron microscopy (TEM) provides extremely detailed high-resolution images of biological structures ranging from subcellular organelles to whole tissues. An advantage of this approach is that TEM allows simultaneous visualization of many different cellular components, providing a holistic view of a cell or tissue, in contrast to fluorescence microscopy which provides spatial info...

Comparison of state-of-the-art atlas-based bone segmentation approaches from brain MR images for MR-only radiation planning and PET/MR attenuation correction

Introduction: Magnetic Resonance (MR) imaging has emerged as a valuable tool in radiation treatment (RT) planning as well as Positron Emission Tomography (PET) imaging owing to its superior soft-tissue contrast. Due to the fact that there is no direct transformation from voxel intensity in MR images into electron density, itchr('39')s crucial to generate a pseudo-CT (Computed Tomography) image ...

Three-dimensional reconstruction of axonemal outer dynein arms in situ by electron tomography.

We present here for the first time a 3D reconstruction of in situ axonemal outer dynein arms. This reconstruction has been obtained by electron tomography applied to a series of tilted images collected from metal replicas of rapidly frozen, cryofractured, and metal-replicated sperm axonemes of the cecidomid dipteran Monarthropalpus flavus. This peculiar axonemal model consists of several microt...
